</p>
<p>Biological processes involving macromolecules and macromolecular assemblies often entail complex structural and positional dynamics of the molecules as important functional elements. These dynamics on the microsecond or longer time scale can be efficiently monitored with single molecule methods. Based on single molecule fluorescence and force measurements, we study tRNA selection by the ribosome, DNA clamp loading, and changes in nucleosome structure and dynamics induced upon DNA methylation and histone acetylation. I will present our findings in the three systems which demonstrate the stochastic nature of slow biological processes engaging macromolecular assemblies.
